Objective Investigate the prevention effects of nabumetone to the damaged esophageal mucosa in the model of reflux esophagitis, and the effect of expression of EGFR,ODC,CDH in the progress of reflux esophagitis. Methods 100 SD rats divided into three groups in random. Group Y: operation + saline as positive controls(46); Group R: operation + nabumetone(46); Group C: normal control(8). The lesions of esophageal mucosa were observed in the 5th, 17th, 28th , 40th week in group Y, R and 40th week in group C respectively. Evaluated the change of EGFR mRNA, ODC mRNA, CDH mRNA in the progress of reflux esophagitis and the effect of nabumetone. Results 1. The lesions of esophageal mucosa in different time and the incidence of Barrett’s esophagus(BE) in group R were lower than group Y(P<0.05). 2. The expression of EGFR mRNA increased gradually from normal→RE→BE→EAC in group Y, and higher than group C. Is higher in group R than in group C, but is lower than group Y in 40th week. 3. The expression of ODC mRNA in group Y were higher than that were in group R and group C in different times(P<0.05).4. the expression of CDH mRNA were lower in group Y, and there was significant difference between group Y and R, C in 28th , 40th week (P<0.05).Conclusion In the progress of RE, EGFR mRNA, ODC mRNA increased gradually, CDH mRNA decreased gradually. This suggests that enhanced expression of EGFR mRNA, ODC mRNA and decreased expression of CHD mRNA may occur early during Barrett's associated neoplastic transformation. Nabumetone reduced mucosa injury in reflux esophagitis and against the development of BE by effect the expression of EGFR, ODC, CDH. |
