E2F转录因子家族的研究进展

摘要:

目的:总结国内外对于E2F转录因子家族的研究现状,探讨该转录因子家族在细胞周期调控、细胞凋亡以及肿瘤发生发展等方面的作用机制、研究意义及应用前景。方法:应用CNKI及PubMed全文数据库检索系统,以“E2F转录因子家族、细胞周期、细胞凋亡、肿瘤”等为关键词,检索1986-2012年的相关文献。纳入标准:1)E2F转录因子家族相关研究背景;2)E2F转录因子家族与细胞凋亡;3)E2F家族对细胞周期的调控;4)E2F家族在肿瘤中的表达及意义。符合要求并纳入分析的文献38篇。结果: E2F家族作为一个庞大的转录因子家族,在细胞周期的调控和细胞凋亡中具有重要的作用,并且在不同的肿瘤中具有抑癌基因和促癌基因的双重功能。结论:E2F家族因其自身功能的复杂性及广泛性,在今后肿瘤的基因治疗方面,具有很广阔的应用前景,或许能为不同种类的肿瘤治疗提供新的治疗靶点。

Objective: To review the domestic and abroad articles related to the research status of E2F transcription factors, then discuss the mechanism of these factors on cell cycle, apoptosis and tumor progression.Methods:“ E2F transcription factors,cell cycle,apoptosis, neoplasm”were searched as key words by CNKI and PubMed series full text database retrieval system from 1986 to 2012. Choice criteria :1) the research background of E2F transcription factors; 2) E2F transcription factors and apoptosis; 3)the regulate and control of E2F transcription factors during cell cycle ;4) the expression and meaning of E2F transcription factors during tumor progression. According to the choice criteria, 38 papers were finally selected and analyzed. Results: E2F transcription factors play a key role in cell cycle control and apoptosis. E2Fs can also function as both oncogene and tumor suppressor gene during tumor progression. Conclusion: E2F transcription factors have very broad application prospects due to the complexity of their own function, and maybe can offer new therapeutic targets for different kinds of tumor treatment in tumor gene therapy in the future.
