两种人外周血单核细胞来源树突状细胞表型及功能比较

摘要:

目的 探索体外培养的干扰素-树突状细胞(IFN-ÐC)和白介素4-树突状细胞(IL-4-ÐC)在表型及功能上的区别。 方法 人外周血单核细胞在体外分别用细胞因子GM-CSF联合IFN-α或者GM-CSF联合IL-4诱导培养为IFN-ÐC或IL-4-ÐC,随后加入ÞNF-α促进ÐC成熟,流式细胞术检测两种ÐC的表型;两种ÐC负载CMVpp65蛋白后刺激自体Þ淋巴细胞激活,细胞内染色方法检测分泌IFN-γ的Þ细胞的比例。结果 IFN-ÐC和IL-4-ÐC均具有典型的树突状细胞形态和免疫表型,但IFN-ÐC表达更高水平的CÐ80、CÐ8³、CÐ86;负载CMV pp65蛋白的不成熟IFN-ÐC及不成熟IL-4-ÐC能够分别诱导(0.²8±0.07)%及(0.¹²±0.0²)%的自体CÐ8+ Þ细胞激活(P<0.05),成熟IFN-ÐC及成熟IL-4-ÐC能够分别激活(¹.0³±0.²¹)%及(0.³5±0.¹4)%的自体CÐ8+ Þ细胞激活(P<0.05)。结论 与IL-4-ÐC相比,IFN-ÐC表达更高水平的CÐ80、CÐ8³、CÐ86,IFN-ÐC负载CMV pp65蛋白能够更有效激活人抗原特异性CÐ8+ Þ细胞。
