组蛋白脱乙酰酶11抑制p53在垂体肿瘤细胞中的表达

摘要:

目的:探讨组蛋白脱乙酰酶11(HDAC11)调节垂体瘤中细胞凋亡的作用机制。方法:选取我院2015年1月至2016年12月收集的垂体瘤标本50例,同时选取距离肿瘤边缘>2 cm的癌旁组织作为对照。所有标本均经病理诊断证实,术前患者未经放疗、化疗。SYBR green实时荧光定量PCR法检测不同亚型组HDAC11的相对表达量,并分析其与p53的相关性以及相互作用机制。结果:(1) HDAC11与垂体瘤细胞中p53的表达呈负相关;(2) HDAC11在垂体瘤细胞中与HEY1形成了一个复合体;(3) HDAC11可以使垂体瘤细胞中HEY1去乙酰化;(4) HDAC11抑制垂体瘤细胞中p53的表达。结论:垂体瘤细胞中高表达HDAC11,而HDAC11可以抑制p53在垂体肿瘤细胞中的表达,HDAC11在垂体瘤的发生发展中起着重要作用。

Objective: To research the role of Histone deacetylase 11(HDAC 11) in the interference with apoptosis in pituitary tumor.Methods: 50 cases of pituitary tumor collected from our hospital in 2015-2016 were selected and the adjacent tissueswith the edge>2 cm were selected as the control. All specimens were confirmed by pathological diagnosis, without preoperative radiotherapy, chemotherapy. The relative expression of HDAC 11 in pituitary tumor cell and adjacent tissues was analyzed by SYBR green real-time quantitative PCR. The relative expression of HDAC 11 in pituitary tumor cells was analyzed and the relationship between HDAC11 and p53 was analyzed.Results: (1) HDAC11 was negatively correlated with p53 in pituitary tumor cells;(2) HDAC11 formed a complex with HEY1 in pituitary tumor cells;(3) HDAC11 modulated the acetylation status of HEY1 in pituitary tumor cells;(4) HDAC11 prevented p53 expression in pituitary tumor cells.Conclusion: HDAC11 can inhibit the expression of p53 in pituitary tumor cells, and HDAC11 plays an important role in the development and progression of pituitary tumor.
