Beclin-1和bcl-2蛋白对前列腺癌的发生、进展的作用

摘要:

目的:探讨前列腺癌组织beclin-1和bcl-2蛋白表达及临床意义。方法:采用免疫组织化学染色检测75例前列腺癌患者(PC组)、40例前列腺高级别上皮内瘤变患者(HGPIN组)和40例前列腺增生患者(BPH组)组织beclin-1、bcl-2蛋白表达,比较beclin-1、bcl-2与前列腺癌患者临床资料的关系。结果:PC组beclin-1蛋白阳性表达率为54.7%(41/75),HGPIN组为67.5%(27/40),BPH组为92.5%(37/40),PC组beclin-1蛋白阳性表达率明显低于HGPIN组和BPH组,差异具有统计学意义(P < 0.05)。PC组bcl-2蛋白阳性表达率为85.4%(64/75),HGPIN组为60.0%(24/40),BPH组为32.5%(13/40),PC组bcl-2蛋白阳性表达率明显高于HGPIN组和BPH组,差异具有统计学意义(P < 0.05)。Beclin-1、bcl-2表达与前列腺癌患者年龄、病理分级、前列腺炎病史、淋巴结转移等无关(P>0.05),前列腺癌Gleason评分越高,beclin-1阳性率越低,bcl-2阳性率越高,差异具有统计学意义(P < 0.05);TNM分期越高,beclin-1阳性率越低,bcl-2阳性率越高,差异具有统计学意义(P < 0.05)。前列腺癌组织beclin-1表达与bcl-2呈显著负相关(r=-0.241,P < 0.05)。结论:前列腺癌中beclin-1基因缺失,bcl-2基因高表达,beclin-1/bcl-2共同参与了前列腺癌的发生和进展。

Objective: To explore the expression of beclin-1 and bcl-2 in prostate cancer and its clinical significance. Methods: The protein expression of beclin-1, bcl-2 was measured by immunohistoehemistry assay in seventy-five prostate cancer tissues, forty high grade prostatic intraepithelial neoplasia tissues and forty benign prostatic hyperplasia tissues, The relationship between beclin-1, bcl-2 and prostate cancer patients clinical data were compared. Results: The positive rate of beclin-1 protein in the PC group was 54.7%(41/75), 67.5%(27/40) in the HGPIN group and 92.5%(37/40) in the BPH group, the positive rate of beclin-1 protein in the PC group was significantly lower than that in the HGPIN group and BPH group, and HGPIN group was significantly lower than that in the BPH group, the difference was statistically significant(P<0.05). The positive rate of bcl-2 protein in the PC group was 85.4%(64/75), 60.0%(24/40) in the HGPIN group and 32.5%(13/40) in the BPH group, the positive rate of bcl-2 protein in the PC group was significantly higher than that in the HGPIN group and BPH group, and HGPIN group was significantly higher than that in the BPH group, the difference was statistically significant(P<0.05). There was no significant difference between beclin-1, bcl-2 and patients age, pathological grade, prostatitis history and lymph node metastasis(P>0.05), the score of Gleason in prostate cancer was higher, the positive rate of beclin-1 was weaker, and the positive rate of bcl-2 was stronger, the difference was statistically significant(P<0.05). The grade of TNM stage in prostate cancer was higher, the positive rate of beclin-1 was weaker, and the positive rate of bcl-2 was stronger, the difference was statistically significant(P<0.05). Beclin-1 expression was found to have negative correlation with bcl-2 in prostate cancer(r=-0.241, P < 0.05). Conclusion: Down-expression of beclin-1 gene and over-expression of bcl-2 gene exit in prostate cancer, beclin-1/bcl-2 may contribute to regulate the tumorigenesis and development of PC cells.
