AhR蛋白的表达水平与胚胎停育的相关性研究

摘要:

目的: 探究并分析芳香烃受体(AhR)蛋白表达水平与胚胎停育的相关性。方法: 选取2016年6月-2017年6月收治的胚胎停育孕妇50例,作为观察组;同时选取未发生胚胎停育的人流孕妇50例,作为对照组。观察并记录两组孕妇外周血及绒毛组织中AhR表达情况,并对影响胚胎停育的相关因素作单因素方差分析以及Logistic回归分析,分析各因素与胚胎停育的相关性。方法: 两组间比较,观察组外周血及绒毛组织中AhR表达水平明显高于对照组(P < 0.05);单因素方差分析结果表明:具有体外受精-胚胎移植病史、盆腔炎病史、人工流产病史的产妇其胚胎停育发生率显著增高,差异具有统计学意义(P < 0.05);同时,外周血AhR蛋白的表达量(≥ 4 ng·ml-1)其胚胎停育的发生率显著增高,差异有统计学意义(P < 0.01);绒毛组织AhR mRNA的相对表达量 ≥ 2,其胚胎停育的发生率显著增高,差异有统计学意义(P < 0.01);绒毛组织AhR蛋白的相对表达量 ≥ 0.7,其胚胎停育的发生率显著增高,差异具有统计学意义(P < 0.01)。Logistic回归分析得出盆腔炎病史、人工流产病史、外周血AhR蛋白、绒毛组织AhR mRNA、绒毛组织AhR蛋白与胚胎停育的发生具有相关性(P<0.05)。方法: 盆腔炎病史、人流病史、外周血AhR蛋白水平、绒毛组织AhR mRNA水平、绒毛组织AhR蛋白水平与胚胎停育的发生具有相关性,可为临床诊断胚胎停育提供参考价值。

Objective: To explore and analyze the correlation between the expression level of aromatic hydrocarbon receptor (AhR) protein and embryonic diapause. Methods: From June 2016 to June 2017, 50 pregnant women with embryonic diapause were selected as observation group, at the same time, 50 pregnant women who did not have embryonic diapause were selected into control group. The expression of AhR in peripheral blood and chorionic villi of the two groups was observed and recorded, and the related factors affecting embryo sterilization were analyzed by univariate variance analysis and Logistic regression analysis, and the correlation between each factor and embryo diapauce was analyzed. Results: Compared with the control group, the expression of AhR in the peripheral blood and villus tissues of the observation group was significantly higher than that of the control group (P<0.05). The results of univariate variance analysis showed that the in vitro fertilization-embryo transfer history, the history of pelvic inflammatory disease, and the history of abortion were significantly different for the incidence of embryonic diapause(P<0.05). At the same time, the incidence of embryonic diapause of different concentrations of peripheral blood AhR protein and different expression of villous tissue AhR mRNA and protein was significantly different (P<0.05); Logistic regression analysis showed that there was a correlation between the history of pelvic inflammation, induced abortion, AhR protein in peripheral blood and AhR mRNA, chorionic villi AhR protein in chorionic villi (P<0.05). Conclusion: The history of pelvic inflammatory disease, the history of abortion, the AhR protein of peripheral blood, the AhR mRNA of villus tissue, and the AhR protein of villous tissue are significantly correlated with the embryonic diapause, which can provide reference value for clinical diagnosis of embryonic diapause.
