LncRNA HCP5通过调节has-miR-600/HMGB1轴影响非小细胞肺癌细胞A549的增殖和侵袭

摘要:

目的: 探讨LncRNA HCP5调节has-miR-600/HMGB1轴对非小细胞肺癌(NSCLC) A549细胞的影响机制。方法: TCGA数据库配对NSCLC病人肿瘤组织和癌旁组织的基因表达;GEO数据库筛选差异表达的miRNA;质粒构建和转染A549细胞过表达has-miR-600或过表达LncRNA HCP5,噻唑蓝(MTT)法检测细胞增殖,细胞迁移实验检测细胞侵袭能力。结果: 肿瘤组织中高迁移率族蛋白B1(HMGB1)表达量为(13.6±1.38),显著高于癌旁组织的(12.8±1.27),差异有统计学意义(t=2.444,P=0.040);A549细胞中过表达has-miR-600降低HMGB1的表达(t=2.859,P=0.021),降低细胞的增殖和侵袭能力(t=2.571,P=0.033;t=2.418,P=0.042);A549细胞中过表达LncRNA HCP5增加HMGB1蛋白水平(t=2.446,P=0.040),细胞侵袭能力增强(t=2.389,P=0.044);而has-miR-600与LncRNA HCP5共转染则减弱LncRNA HCP5过表达的影响。结论: LncRNA HCP5调控has-miR-600/HMGB1轴影响非小细胞肺癌A549细胞的增殖和侵袭,可为NSCLC的治疗提供一个潜在的新靶点。
