LncRNA SOX2OT、miR-211在非小细胞肺癌患者组织中的表达及其预后相关性

摘要:

目的:检测长链非编码RNA SOX2重叠转录本(LncRNA SOX2OT)、微小RNA-211(miR-211)在非小细胞肺癌(NSCLC)患者组织中的表达水平,并探究其预后相关性。方法:选取2014年1月至2016年1月于本院住院治疗的85例NSCLC患者癌组织作为NSCLC组,选取其相应癌旁正常组织作为对照组。采用实时荧光定量PCR (qRT-PCR)法检测组织LncRNA SOX2OT、miR-211水平;Pearson法分析LncRNA SOX2OT、miR-211的相关性;分析NSCLC组织LncRNA SOX2OT、miR-211水平与患者临床病理特征的关系;Kaplan-Meier法分析NSCLC组织LncRNA SOX2OT、miR-211表达与患者生存率的关系;多因素Cox回归分析影响NSCLC患者预后的因素。结果:NSCLC组LncRNA SOX2OT表达水平高于对照组,miR-211表达水平低于对照组,差异具有统计学意义(P<0.05)。NSCLC患者癌组织中LncRNA SOX2OT与miR-211表达水平呈负相关(r=-0.501,P<0.05)。LncRNA SOX2OT、miR-211表达与年龄、性别无关,差异无统计学意义(P>0.05);与组织学类型、淋巴结转移、分化程度、TNM分期有关,差异有统计学意义(P<0.05)。LncRNA SOX2OT低表达组5年内存活率(85.00%)高于高表达组(24.44%);miR-211低表达组5年内存活率(24.39%)低于高表达组(79.55%),差异有统计学意义(χ2=31.169、25.915,P<0.05)。LncRNA SOX2OT高表达、miR-211低表达、TNM Ⅲ+Ⅳ期是影响NSCLC患者不良预后的危险因素(P<0.05)。结论:LncRNA SOX2OT、miR-211可能存在协同作用,共同参与NSCLC的发生、发展,两者对NSCLC患者的病情评价及治疗可能有重要意义。

Objective: To detect the expression levels of long non-coding RNA SOX2 overlapping transcript(LncRNA SOX2OT) and microRNA-211(miR-211) in non-small cell lung cancer(NSCLC) tissues, and to explore their prognostic relevance.Methods: The cancer tissue from 85 cases of NSCLC patients who were hospitalized in our hospital from January 2014 to January 2016 were selected as NSCLC group, and their corresponding normal tissues were selected as control group. Real-time fluorescence quantitative PCR(qRT-PCR) was used to detected the levels of LncRNA SOX2OT and miR-211; Pearson method was used to analyze the correlation between LncRNA SOX2OT and miR-211; the relationship between the levels of LncRNA SOX2OT and miR-211 in NSCLC tissues and clinicopathological characteristics was analyzed; Kaplan-Meier method was used to analyze the relationship between the expression of LncRNA SOX2OT and miR-211 and the survival rate of NSCLC tissue; multivariate Cox regression analysis was used to analyze the prognostic factors of NSCLC patients.Results: The expression level of LncRNA SOX2OT in the NSCLC group was higher than that in the control group, and the expression level of miR-211 was lower than that in the control group(P<0.05). There was a negative correlation between the expression levels of LncRNA SOX2OT and miR-211 in cancer tissues of NSCLC patients(r=-0.501, P<0.05). The expression of LncRNA SOX2OT and miR-211 was not related to age and gender(P>0.05); but was related to histological type, lymph node metastasis, differentiation degree and TNM stage(P<0.05). The 5-year survival rate of LncRNA SOX2OT low expression group(85.00%) was higher than that of high expression group(24.44%); the 5-year survival rate of miR-211 low expression group(24.39%) was lower than that of high expression group(79.55%), the differences were statistically significant(χ2=31.169, 25.915,P<0.05). High expression of LncRNA SOX2OT, low expression of miR-211 and TNM stage Ⅲ+Ⅳ were risk factors for poor prognosis of NSCLC patients(P<0.05). Conclusion: LncRNA SOX2OT and miR-211 may have synergistic effect, and play a role in the occurrence and development of NSCLC. They may be of great significance in the evaluation and treatment of NSCLC patients.
