小细胞肺癌发生和预后相关血清外泌体分子标记物的鉴定和验证

摘要:

目的: 基于血清外泌体数据筛选小细胞肺癌患者核心分子标记物,用于判断小细胞肺癌的发生和预后。方法: 从血清外泌体数据库exoRBase中下载健康人群和小细胞肺癌患者基因表达数据集(检索起止: 建库至2021年12月10日),采用加权基因共表达网络分析对由118例健康人群和36例小细胞肺癌患者组成的数据集进行分析,通过蛋白-蛋白相互作用以及MOCDE等多种算法筛选核心分子并利用外部ONCOMINE和GEO数据库中多个数据集进行外部验证。结果: 基因共表达网络分析(weighted gene co-expression network,WGCNA)筛选出红色模块与小细胞肺癌发生密切相关,利用蛋白-蛋白相互作用网络等多种算法对模块中基因深入分析,筛选出H2BFS基因作为核心分子。并通过内部数据集验证H2BFS表达与小细胞肺癌发生相关,可以作为有效的血清外泌体分子标记物。同时,外部ONCOMINE证实H2BFS在肺癌中高表达,且在多个GEO数据集(GSE377450、GSE31210和GSE29013)中验证高表达H2BFS患者预后更差。结论: 利用血清外泌体数据库并结合WGCNA算法,筛选获得的H2BFS基因可以作为小细胞肺癌患者有效的血清外泌体分子标记物。

Objective: To screen out the effective biomarkers in small cell lung cancer(SCLC) patients based on serum exosome data.Methods: Gene expression of healthy people and SCLC were downloaded from serum exosome database(Time periods:up to 2021.12.10). Weighted gene co-expression network analysis(WGCNA) was used to analyze the data. Core molecules were screened and external validation was performed using ONCOMINE and GEO databases.Results: The red module screened by WGCNA was related to the occurrence of SCLC. The H2BFS gene was selected as the core molecule by using protein-protein interaction network and other algorithms. The expression of H2BFS was correlated with the occurrence of SCLCand could be used as an effective molecular marker of serum exosome. At the same time, the external ONCOMINE confirmed that H2BFS was highly expressed in lung cancer, and in multiple GEO data sets:GSE377450, GSE31210 and GSE29013, patients with high H2BFS expression had worse prognosis. These results suggested that H2BFS could be used as a biomarker for serum exosomes in patients with SCLC.Conclusion: Using serum exosome database combined with WGCNA, the H2BFS gene can be used as an effective serum exosome molecular marker in patients with SCLC.
