树突状细胞在类风湿关节炎免疫耐受治疗中的作用

类风湿关节炎(rheumatoid arthritis,RA)是常见的自身免疫性疾病之一,其发生和发展是由多种诱发因素和机体免疫异常所造成的,其中抗原提呈功能的异常是一个重要方面。树突状细胞(dendritic cells,DC)是目前所知的机体内功能最强的抗原提呈细胞(APC),它不仅是机体免疫应答的始动者,而且在诱导免疫耐受、调节T细胞介导的免疫应答方面都发挥着重要作用。国内对于DC在RA方面的研究也越来越受到关注,该研究也为RA的治疗提供了新的治疗思路。1 DC的来源及其生物学特性DC因其表面有许多树枝状突起而得名,于1973年首次由Steinman和Cohn发现。DC均来源于体内的多能造血干细胞,主要分为三大类:(1)皮肤DC,即郎格罕细胞(Langerhans cell,LC);(2)髓系来源的DC(myeloid-drived DC),与单核细胞、粒细胞有共同的祖先;(3)淋巴系来源的DC(lymphoidr-elated DC),与T细胞、B细胞有共同的前体细胞。其中髓系DC是体内最主要的DC。大多数DC来源于骨髓,由骨髓进入外周血,再分布到全身各组织。DC广泛分布于除脑以外的全身各脏器,数量极...
